Through our comprehensive approach, the Basser Center for BRCA at Penn Medicine’s Abramson Cancer Center has set a global standard for BRCA-related research, education, and patient care. We remain deeply committed to transforming the future for BRCA mutation carriers by driving innovative research at Penn and around the world to accelerate scientific breakthroughs.

This report is not a complete account of everything we’ve accomplished over the past year, but rather a spotlight on several key areas where Basser is making a significant impact. You’ll read about our efforts to improve early detection and harness the potential of cancer interception, our growing focus on research for male BRCA mutation carriers, and how we’re helping to build a strong pipeline of future leaders in the field.

Unlocking Early Detection & Interception

Established in 2022 by Basser founders Mindy and Jon Gray, the Basser Cancer Interception Institute (BCII) is working to dramatically disrupt the timeline of cancer treatment, improving early detection and intercepting disease at its earliest stages, when the first abnormal cells begin to develop. Basser researchers are tackling cancer interception from multiple angles. The highlights below showcase some of the most groundbreaking aspects of this work.

Leveraging the Immune System to Fight Cancer

Work continues on Basser’s pioneering study of a new cancer vaccine for individuals with a BRCA1 or BRCA2 mutation, led by Susan Domchek, MD. Enrollment has closed for the latest cohort, which included 28 healthy BRCA mutation carriers with no cancer history and 16 BRCA mutation carriers with a prior cancer diagnosis.

Dr. Domchek and her team will monitor participants over the next two years to assess safety and determine whether their T cells are successfully recognizing telomerase—one of the vaccine’s targets—which plays a role in 95% of human cancers.

Studying the Origins of BRCA-Related Breast Cancer

Emerging research by Carman Li, PhD, a new Basser investigator, suggests that cancer predisposition cannot be fully explained by the prevailing “two-hit” hypothesis, which posits that two “bad copies” of a gene are necessary for cancer to develop. She has developed the first mouse models genetically engineered with a single mutated copy of the BRCA gene and is studying epigenetic changes to better understand cancer risk and progression. Though still in its early stages, this research has the potential to improve risk stratification for mutation carriers and identify new targets for cancer interception.

Pancreatic Cancer Interception

Cancer interception aims to eliminate early “pre-malignant” lesions before they develop into full-blown cancer. This approach may be especially important for individuals who face an increased risk of BRCA-related pancreatic cancer—a disease for which early detection methods are currently lacking.

With support from the Becker Family, Robert H. Vonderheide, MD DPhil and Ben Stanger, MD, PhD are examining the oncogene KRAS, the root acquired cause of most cases of pancreatic cancer, as a potential method for interception. Early results in the laboratory are promising, and a clinical trial has been designed.

New Collaborative Interception Initiative

The Basser Immune-Interception Initiative is a transformative new approach that unites experts in cancer biology, immunology, and RNA therapeutics from the Perelman School of Medicine, Abramson Cancer Center, School of Veterinary Medicine, and the Penn Institute for RNA Innovation. Led by Christopher Hunter, PhD at the School of Veterinary Medicine, the goal is to generate and test therapies designed to elicit immune responses specifically against early tumor-associated antigens. It will provide a platform for development, advancing the most promising basic science research findings into a preclinical screening pipeline, with the end goal of developing new, personalized cancer interception strategies for clinical trials.

Improving Ovarian Cancer Early Detection

Prior research by Ronny Drapkin, MD, PhD identified the protein ORF1p as a potential biomarker for high-grade serous ovarian cancer. Building on this discovery, Dr. Drapkin and his team have developed an ultrasensitive assay capable of detecting minute amounts of ORF1p in the blood and are exploring its potential as a vaccine target.

Working in collaboration with Victor E. Velculescu, MD, PhD, of Johns Hopkins Kimmel Cancer Center, Dr. Drapkin is also working to enhance early detection through AI-driven liquid biopsies. This approach combines AI with two established protein biomarkers, CA-125 and HE4. These research efforts represent a significant step toward developing a reliable screening tool for ovarian cancer, ultimately saving countless lives.

A Focus on Men & BRCA

Established in 2022 through a generous gift from Julie and Martin Franklin, the Men & BRCA Program—led by Kara Maxwell, MD, PhD—continues to make a significant impact by raising awareness of BRCA mutations in men and advancing early detection, prevention, and treatment of BRCA-related cancers in men.

Developing a Standard of Care for Male BRCA Mutation Carriers

Kara Maxwell, MD, PhD, in collaboration with Heather H. Cheng, MD, PhD of Fred Hutch Cancer Center, published BRCA1, BRCA2 and Associated Cancer Risks and Management for Males in JAMA Oncology. This peer-reviewed paper brings together various approved guidelines into a single, accessible resource for clinicians. It represents the first comprehensive clinical resource focused specifically on BRCA in men—addressing a critical gap in care for physicians treating male BRCA mutation carriers.

Understanding Male Breast Cancer

Jennifer Q. Zhang, MD, is studying tumor samples to better understand the underlying biology of male breast cancer, with a particular focus on how it differs from female breast cancer. Her findings will help guide the development of more targeted and effective therapies specifically for men with breast cancer.

Improving Early Detection for Prostate Cancer

Daniel J. Lee, MS, MD leads the PATROL study, aimed at improving early detection of prostate cancer in men with a BRCA mutation. The study uses lower PSA thresholds than the current standard as triggers for biopsy. The trial has already identified cancers in this high-risk population and may serve as a model to enhance screening and early detection.

Basser Scientific Symposium

Each year, the Basser Scientific Symposium brings together more than 500 BRCA-focused physicians, scientists, genetic counselors, and care providers from around the world to share their work and explore recent advances and future directions in BRCA research and clinical care.


A highlight of the symposium is the Basser Global Prize, established by Shari and Len Potter. This year, the prize was awarded to Patrick Sung, DPhil, of the University of Texas Health Science Center at San Antonio.

Pearl & Philip Basser Innovation Research Award

This research award, established and endowed by the late Pearl and Philip B. Basser, honors an early career investigator conducting innovative BRCA-related research. The 2025 award was presented to Vivek K. Narayan, MD, MSCE, in recognition of his BRCA-related prostate cancer research.

Supporting the BRCA Leaders of Tomorrow

Through several key initiatives, Basser is helping to develop the next generation of leaders in genetics and BRCA-related science—paving the way for future breakthroughs in research and care.


Postdoctoral Fellowship Program

The newly launched Postdoctoral Fellowship Program aims to train investigators across a range of disciplines—including basic and translational science, implementation and behavioral science, preventive medicine, bioinformatics, and clinical trials. Fellows will work alongside a Basser Core Investigator and receive funding to advance their independent research.

By supporting postdoctoral and clinical fellows during this formative stage of their careers, the program will help establish them in diverse areas of BRCA1/2-related research. This effort will strengthen the Basser Center’s research enterprise and ensure a growing network of experts at Penn and institutions across the country, all dedicated to ending BRCA-related cancers.

Basser Summer Scholars

The Summer Scholars Internship Program is an eight-week intensive research experience for undergraduate students interested in genetics. Participants are immersed in laboratory, classroom, and clinical research settings, gaining exposure to basic science, cancer genetics, and genetic counseling—all under the guidance of Basser Center leaders.

Now in its fifth year, the program hosted nine students in summer 2025.


Genetic Counseling Scholarship

Basser is committed to expanding the pipeline of genetic counselors to ensure that all BRCA mutation carriers have access to critical information about their mutation status. For many aspiring counselors, the cost of obtaining a graduate degree in genetic counseling poses a significant barrier to entering the field.

To help overcome this challenge, the Basser Genetic Counseling Scholarship was established in 2021 by Basser Center Advisory Board members Angela and the late Joe Campolo, Cindy and Brian Gavin, Michele and Kevah Konner, Heidi and Richard Rieger, Mara Burros-Sandler, and Helene and Rob Sorin. The scholarship provides full tuition support to a student pursuing a Master’s in Genetic Counseling at Penn’s Perelman School of Medicine.

Genetic Counselors

Basser Genetic Counselors provide essential care for patients interested in understanding their hereditary risk. Additionally, they conduct important research to improve access to genetic counseling and testing, helping to ensure that everyone has access to this life-saving information.

Advisory Board

The Basser Center Advisory Board (BCAB) advocates, informs, and engages with the broader community on behalf of the Basser Center. The Advisory Board plays a vital role in advancing and supporting the Center’s mission to accelerate BRCA-related research, care, and education.

Young Leadership Council

The Young Leadership Council (YLC) includes more than 200 members from around the world who help disseminate information on BRCA-related research and advances. Through informative webinars and panel discussions, they play an active role in educating the broader community. In addition to outreach and education, the YLC raises funds and awards research grants to support early career investigators advancing BRCA-related science. YLC Young Investigator Award

Each year, the Basser Young Leadership Council raises funds to support early career investigators conducting BRCA-related research. In 2024, the YLC awarded two research grants: one to Anne Marie McCarthy, ScM, PhD, for her work on BRCA-related breast cancer; and another to Carman Li, PhD, for her research on BRCA-related cancer biology.

In October 2024, the Basser Center’s Young Leadership Council hosted An Evening of Storytelling, an unforgettable night of connection, community, and captivating stories of lives impacted by BRCA mutations.

Parents Leadership Community

The Parents Leadership Community (PLC) is a group of parents supporting their children who are, or may be, affected by a BRCA1 or BRCA2 mutation. The PLC serves as both a supportive forum for parents and a valuable resource for staying informed about the latest advances in BRCA research, care, and risk management. In April 2024, the Parents Leadership Community and Young Leadership Council presented a Basser Pearls of Wisdom conversation featuring Susan Domchek, MD, Lola Fayanju, MD, MA, MPHS, and Jessica Fayne Rose, DO. This informative and supportive discussion focuses on mastectomy and reconstruction — covering what to expect before, during, and after surgery; different types of breast reconstruction and implant options; and tips for managing recovery and self-care.

Basser at TODAY

Jill MartinTODAY Show contributor and Basser Center Advisory Board member—helped kick off Breast Cancer Awareness Month on TODAY alongside Dr. Domchek. They highlighted the importance of understanding BRCA mutations and encouraged both men and women to learn more about their family health history.

Basser on Eyewitness News

Dr. Domchek continued her press tour in New York City, speaking with breast cancer survivor and Basser Advisory Board member Stacey Sager on ABC7's Eyewitness News. She shared that the future of breast cancer treatment is brighter than ever, with a wealth of new therapies and research making it into the clinic.



Spreading BRCA Awareness

Basser partnered the Warrior Mensch Foundation, Park Avenue Synagogue, and Central Synagogue to raise awareness of BRCA mutations in the Ashkenazi Jewish population. This initiative included access to genetic testing kits provided by the foundation and an informational webinar hosted by Basser. The effort was supported by the Becker Family in memory of Marc Becker.
